It is recommended to use a polyester sponge with a PP (polypropylene) rod as a sterile test piece for sample collection.
(1) The method of collecting oropharyngeal test specimens: the person's head is slightly inclined, and the mouth is opened wide, exposing the tonsils on both sides. Put the swab across the base of the tongue and wipe the tonsils back and forth with slight force on both sides of the collected person at least 3 times, and then wipe up and down the posterior pharyngeal wall at least 3 times.
(2) The method of collecting nasopharyngeal test specimens: the sampler gently supports the head of the person to be collected with one hand, holds the swab in the other hand, and sticks the swab into the nostril, and then slowly goes deep along the bottom of the lower nasal passage. Arc shape, do not use excessive force to avoid traumatic bleeding. When the tip of the swab reaches the back wall of the nasopharyngeal cavity, gently rotate it once (in case of reflex cough, stay for a while), and then slowly take out the swab.
(3) Sample processing: The collected sample should be processed with the sample buffer provided by this kit as soon as possible (if it cannot be processed immediately, the sample should be stored in a dry, sterilized and strictly sealed plastic tube), at 2℃～8℃ Keep for no more than 24h; long-term storage at -70℃, but repeated freezing and thawing should be avoided.
Inspection Steps
Please restore all reagents to room temperature before testing, and the test shall be conducted at room temperature.
一,Extraction of specimens (see Figure 1)
1. Add 400μL vertically into the sample extraction tube (about 10 drops) of sample buffer solution, then insert the sample into the solution of sample extraction tube, rotate it close to the inner wall for about 10 times, so that the sample can be dissolved in the solution as much as possible.
2. squeeze the cotton swabs along the inner wall of the extraction pipe to keep the liquid in the pipe as far as possible, take out and discard the test piece.
3. cover the drip head.
二,Testing procedures (see Figure 2)
1. Take out the test card from the sealed bag.
2. Add 2 drops (about 80μL) of the processed sample extract to the sample hole of the test card, and then start the timer.
3. Read the result when the test card is placed at room temperature for 15 minutes. Reading results after 20 minutes is invalid
Detailed explanation of test results
The results of the test card are as follows:
① invalid results: there is no reaction line in the quality control line (line C), and the test is invalid, so the experiment should be repeated.
② Negative results: a red color band, quality control line (C line) color.
③ Positive results: two red bands, test line (t line) and quality control line (C line) were colored.
